Conversion of AFLP marker to sequence specific PCR marker in S. leprosula
ABSTRACT
Genetic marker can efficiently obtained by using amplified fragment length polymorphism (AFLP) fingerprinting. One AFLP fragment has been identified specific for S. leprosula. The AFLP marker was isolated from a gel, re-amplified, cloned and sequenced. Primer set was designed from these sequences and converted into a SCAR (Sequence Characterized Amplified Region) marker. The SCAR was used to screen samples of. S.leprosula from 14 widely separated populations in Indonesia. The marker was successfully applied to confirm the species S. leprosula.
