ANALISIS PERUBAHAN FISIOLOGI DAN BIOKIMIA BENIH TENGKAWANG SELAMA PENGERINGAN = Analysis of Physiological and Biochemical of Tengkawang Seeds During Dessication
The research aimed to study physiological and biochemical aspects of Shorea stenoptera seeds deterioration during desiccation process. Based on seeds initial moisture content (46.06%), the target of moisture content in the drying process were 45%, 40%, 35%, 25%, 15%, and 8%. The research used a completely randomized design with one factor: the drying. Each treatment consists of 25 seeds and three replications. The observed physiological parameters were germination percentage, germination rate and germination value, and the biochemical parameters were the content of starch, fat, protein and electrical conductivity. Results showed that the seed viability decreased with the decreasing moisture content. Maximum viability was achieved in fresh seeds (NR: 46.06%) with 85.33% germination, growth rates of 2.65% per day and germination value of 0:43. Biochemical content (fat, starch, and electric conductivity) of seeds of S. stenoptera tended to increase, while the protein tended to decrease with decreasing levels of seeds moisture content. S. stenoptera seeds critical moisture content was supposed to occurr on the target moisture content of 15%, because at 8% target moisture content, seeds were not germinated (germination percentage: 0%). Based on physiological and biochemical parameters, the seed S. stenoptera can be categorized as recalcitrant seeds. = Penelitian ini bertujuan untuk mempelajari aspek fisiologi dan biokimia kemunduran (penurunan viabilitas) benih tengkawang (S. stenoptera) selama proses pengeringan. Berdasarkan kadar air awal benih (46,06%), target penurunan kadar air (TKA) dalam proses pengeringan adalah 45%, 40%, 35%,
25%, 15% dan 8%. Rancangan yang digunakan merupakan Rancangan Acak Lengkap dengan satu faktor yaitu pengeringan. Setiap perlakuan terdiri dari 25 satuan percobaan yang diulang 3 kali. Parameter fisiologi yang diamati meliputi daya berkecambah, kecepatan berkecambah dan nilai perkecambahan, sedangkan parameter biokimia meliputi kandungan pati, lemak, protein dan daya hantar listrik. Hasil penelitian menunjukkan bahwa viabilitas benih semakin menurun sejalan dengan penurunan kadar air benih. Viabilitas maksimum dicapai pada benih segar (KA: 46,06%) dengan daya berkecambah 85,33%, kecepatan tumbuh 2,65% per etmal dan nilai perkecambahan 0,43. Kandungan biokimia (lemak, pati, dan daya hantar listik) benih S. stenoptera cenderung mengalami peningkatan, sedangkan protein cenderung mengalami penurunan dengan semakin menurunnya tingkat kadar air benih. Kadar air kritis benih S. stenoptera diduga terjadi pada target kadar air (TKA) 15%, karena pada TKA 8% benih sudah tidak mampu berkecambah (DB : 0%). Berdasarkan parameter fisiologi dan biokimia, benih S. stenoptera dapat dikategorikan ke dalam kelompok benih rekalsitran.
